What is methylation?

Methylation is a chemical process your cells run billions of times a day. A methyl group (one carbon, three hydrogens) gets added to DNA, proteins, hormones, and neurotransmitters. It helps regulate gene expression, detoxification, cardiovascular health, and mood.

A genetic methylation test looks at variants in the genes that run this cycle — most notably MTHFR, MTR, MTRR, COMT, CBS, and MTHFD1.

The genes that matter

MTHFR (C677T & A1298C)

MTHFR converts folate into 5-MTHF, the form your body actually uses. The C677T variant can reduce enzyme activity by roughly 30% (heterozygous) to 70% (homozygous). A1298C has a milder effect. Neither is a disease on its own.

MTR & MTRR

MTR recycles homocysteine back to methionine using B12. MTRR keeps MTR active. Variants here interact with your B12 status more than they act alone.

COMT

COMT breaks down dopamine, adrenaline, and estrogen. The Val158Met (rs4680) variant influences how quickly you clear these — often discussed in the context of stress response and caffeine sensitivity.

Do you need a dedicated methylation test?

  • Look at each variant's genotype (e.g., CT, TT).
  • Check the strength of the underlying research — a single small study is not proof.

Is MTHFR a disease?

No. It's a common gene variant — roughly 40% of people carry at least one copy.

Should I take methylfolate?

Only with clinical guidance. High-dose methylfolate can worsen anxiety in some people, especially with certain COMT genotypes. Test, don't guess.

Can I trust 23andMe's raw data?
